چکیده

Abstract Pyrazolo[3,4-b]pyridine is a privileged scaffold found in many small drug molecules that possess wide range of pharmacological properties. Efforts to further develop and exploit synthetic methodologies permit the functionalization this heterocyclic moiety warrant investigation. To end, series novel 1,3-disubstituted pyrazolo[3,4-b]pyridine-3-carboxamide derivatives have been prepared by introducing 3-carboxamide using palladium-catalyzed aminocarbonylation methodology employing CO gas generated ex situ two-chamber reactor (COware®). The functional group tolerance optimized protocol highlighted through synthesis diversely substituted C-3 carboxamide pyrazolo[3,4-b]pyridines excellent yields up 99%.

Palladium-catalyzed cyclization of o-alkynyltrifluoroacetanilides followed by isocyanide insertion: synthesis of 2-substituted 1H-indole-3-carboxamides.

A base-controlled synthesis of 2-substituted secondary and tertiary 1H-indole-3-carboxamides through PdCl(2)-catalyzed cyclization of o-alkynyltrifluoroacetanilides followed by isocyanide insertion has been developed. The reaction proceeds smoothly at ambient temperature using O(2) in air as the sole oxidant of the palladium catalyst.

Palladium-Catalyzed One-Pot Approach to 3-(Diarylmethylene)oxindoles from Propiolamidoaryl Triflate.

3-(Diarylmethylene)oxindoles have been synthesized from propiolamidoaryl triflate utilizing a palladium-catalyzed one-pot reaction consisting of three successive reactions: Sonogashira, Heck, and Suzuki-Miyaura. This method allows for the production of a complex skeleton of 3-(diarylmethylene)oxindole from propiolamidoaryl triflate using a commercially available aryl iodide and arylboronic acid...

An expedient route to substituted furans via olefin cross-metathesis.

The olefin cross-metathesis (CM) reaction is used extensively in organic chemistry and represents a powerful method for the selective synthesis of differentially substituted alkene products. Surprisingly, efforts to integrate this remarkable process into strategies for aromatic and heteroaromatic construction have not been reported. Such structures represent key elements of the majority of smal...
